  • "Cell-specific regulatory networks and cell differentiation, fate and reprogramming"

    This year Prof. Yamanaka was awarded a Nobel prize for his work on cell reprogramming. He showed that mature human cells can be reprogrammed to become pluripotent, i.e. stem cells. Since then others have shown that it is possible to induce one fully mature cell type to differentiate into another.
    Understanding the process of cell differentiation at the molecular level has implications reaching far beyond applications to regenerative medicine including development, evolution, disease and synthetic biology. To this aim scientists around the world have been producing high-throughput experimental data on a massive scale to quantify the molecular state of human and mouse cell types, and for time courses over differentiation. This project is to analyse the data, including public data and also private data not yet publicly available.
    What the experimental data provide, for cell types, tissues or time-points during a cell differentiation, is e.g.: the amount of expression of each gene, epigenetic state, which transcription factors regulate which genes, and which transcription factors interact with each other. This allows us to build networks of regulation in each cell type and at each point in time during a differentiation to understand what determines cell fate. Our own SUPERFAMILY database of structural protein domains, intrinsic protein disorder and functions in genomes provides a framework for understanding the molecular evolution of cell types.
